Levitra (Vardenafil)
Drug UsesCoadministration of ketoconazole (200 mg), a potent inhibitor of CYP 3 A 4, withvardenafil (5 mg) produced a 10fold increase in vardenafil AUC and an increase of 4 times in vardenafil Cmax (see Section 4. 4).
Although no specific interaction studies, it is expected that concomitant useother potent CYP 3 A 4 inhibitors (such as itraconazole or ritonavir) levels occur Vardenafil plasma comparable to those produced by ketoconazole and indinavir. Avoid Concomitant use of vardenafil with potent CYP 3 A 4 inhibitors such as ritonavir, indinavir, itraconazole and ketoconazole orally (see Section 4. 4).
Grapefruit juice is a weak inhibitor of CYP 3 A 4 metabolism in the wall locatedintestinal, therefore, may cause small increases in plasma levels of vardenafil (See Section 4. 4).
The pharmacokinetics of vardenafil (20 mg) was not affected by coadministration with H 2 antagonist, ranitidine (150mgbid), digoxin, warfarin, glibenclamide, alcohol (peakaverage blood alcohol level of 73 mg / dl) or single doses of antacid (hydroxidemagnesium / aluminum hydroxide).
